D016 is an investigational technology. No pharmaceutical product based on D016 has been approved by the U.S. Food and Drug Administration.

Technology Identity

D016 – Recombinant Hyaluronidase Technology

D016 is a recombinantly produced hyaluronidase technology characterized through defined biochemical and analytical methods.

The technology was designed to combine PH20-like and HYAL4-like enzymatic functions within a single recombinant protein and is being investigated in the context of pharmaceutical development.

Recombinant Protein

D016 is produced using recombinant biotechnology and is characterized as a recombinant protein rather than an animal-derived hyaluronidase preparation.

No animal-derived active ingredient source is used for D016.

Designed Dual-Activity Profile

D016 was designed to exhibit both PH20-like and HYAL4-like enzymatic functions within one protein architecture.

This biochemical activity profile is investigated in defined assay systems and experimental models relevant to the respective scientific question.

The designed dual-activity profile does not by itself establish superior biological performance, clinical efficacy or therapeutic benefit.

Defined Enzymatic Activity

Under defined assay conditions, D016 has demonstrated a specific enzymatic activity of approximately ≥1.2 million IU/mg.

Activity values depend on assay methodology, substrate conditions and analytical parameters and should therefore be interpreted within the conditions under which they were generated.

Technology Supporting Pharmaceutical Development

D016 provides the underlying recombinant hyaluronidase technology used within Pantarhin’s pharmaceutical-development framework.

BELACT® is the lead investigational pharmaceutical development program based on D016; D016 and BELACT® should therefore not be interpreted as interchangeable product or program designations.

Technology Access and Rights

Pantarhin’s use of, access to or development activities involving D016 do not by themselves establish ownership of the D016 technology or unrestricted rights to manufacture, supply, license or commercialize D016.

Technology ownership, development rights, supply rights, licensing authority and other commercial rights are determined separately under the applicable corporate, intellectual-property and contractual framework.

Rights and disclosures are defined by agreement – not inferred from access to material or information.

D016 should therefore be understood as a defined recombinant hyaluronidase technology whose biochemical characteristics, scientific evidence and pharmaceutical-development relevance are evaluated separately from questions of program status, regulatory approval and intellectual-property ownership.

Biochemical & Technical Profile

Defined Characteristics. Controlled Interpretation.

D016 is characterized through defined biochemical and analytical methods intended to establish identity, enzymatic activity, purity and other quality-relevant characteristics for the respective stage of development.

Publicly stated values describe selected development characteristics and should be interpreted within the applicable assay method, material configuration and development context.

High Specific Enzymatic Activity

Under defined assay conditions, D016 has demonstrated a specific enzymatic activity of approximately ≥1.2 million IU/mg.

Specific-activity values are assay-dependent and may not be directly comparable with values generated using different substrates, methods, reference standards or analytical conditions.

Application- and Development-Specific Purity

Purity requirements for D016 depend on the intended application and stage of development. Pharmaceutical development is pursued with high-purity material, with selected development specifications targeting approximately ≥99% purity.

The applicable purity criterion, analytical method and release requirement are defined within the respective material and development specification.

Defined Recombinant Protein Format

D016 is a recombinantly produced protein with no animal-derived active ingredient source.

The pharmaceutical-development concept does not rely on PEGylation of the D016 protein.

Analytical Characterization

Development-related analytical work evaluates characteristics such as identity, purity, enzymatic activity, protein integrity and other quality attributes relevant to the respective material and development stage.

Additional analytical controls may be introduced or refined as pharmaceutical development advances and the applicable quality requirements become more defined.

Process-Related Quality Attributes

Pharmaceutical development also requires evaluation and control of process-related impurities and other material attributes according to the manufacturing process, analytical capability and applicable development requirements.

Acceptance criteria and analytical methods are established or refined within the relevant development and quality framework.

Controlled Technical Disclosure

Detailed manufacturing processes, production constructs, host-system information, purification strategy, analytical methods and batch-specific data are not required for the public description of D016 and may be disclosed separately where appropriate within qualified scientific, development or diligence discussions.

Public website information does not replace controlled technical documentation, batch-specific release documentation or project-specific specifications.

The technical profile of D016 therefore evolves together with its development stage: selected biochemical characteristics can be stated publicly, while final specifications, analytical requirements and manufacturing controls remain subject to the applicable development, quality and regulatory framework.

Technical characteristics described on this website do not constitute an approved pharmaceutical specification, a guarantee of batch conformity or evidence of clinical safety, efficacy or therapeutic benefit.

Biological Rationale

Exploring Enzymatic Activity Within Extracellular Matrix Biology

Hyaluronidase-related enzymes interact with glycosaminoglycan components of the extracellular matrix. D016 was designed with PH20-like and HYAL4-like enzymatic functions to investigate a broader biochemical activity profile within this biological context.

This rationale provides a basis for experimental investigation but does not establish that changes observed in biochemical or preclinical systems will translate into a clinically meaningful effect.

Extracellular Matrix Biology

The extracellular matrix is a dynamic biological environment containing structural proteins, glycosaminoglycans and other molecular components that contribute to tissue organization and biological signaling.

D016 is investigated in relation to selected extracellular-matrix glycosaminoglycan substrates relevant to its designed enzymatic activity profile.

PH20-like Enzymatic Function

The PH20-like component of the D016 activity profile is associated with hyaluronidase activity toward hyaluronan-related substrates under defined experimental conditions.

This biochemical activity represents one component of the D016 design and should not be interpreted as evidence of a specific therapeutic effect.

HYAL4-like Enzymatic Function

The HYAL4-like component broadens the designed biochemical profile toward additional glycosaminoglycan-related substrate activity beyond the PH20-like component.

Experimental characterization includes investigation of activity associated with chondroitin-sulfate-related substrates.

Combined Biochemical Profile

The scientific rationale for D016 is not based on simply reproducing one native hyaluronidase function, but on evaluating the biological relevance of a designed recombinant protein exhibiting complementary PH20-like and HYAL4-like enzymatic characteristics.

The presence of complementary enzymatic characteristics does not demonstrate additive, synergistic or superior therapeutic performance.

From Biochemical Activity to Biological Relevance

Biochemical enzyme activity represents the starting point for development. Experimental models are required to determine whether and under which conditions that activity produces biologically relevant effects.

Any observed biological effect must then be evaluated independently for reproducibility, dose relationship, model relevance, safety implications and potential translational significance.

Cardiovascular Development Context

Pantarhin is investigating whether the biochemical and biological characteristics of D016 may have relevance to selected cardiovascular disease processes within the BELACT® development framework.

The scientific rationale does not establish that extracellular-matrix modification will prevent, reverse or treat cardiovascular disease in humans.

The biological rationale is evaluated through biochemical, experimental and preclinical evidence. Those evidence levels and their limitations are described separately on the Science & Evidence page.

Explore Science & Evidence →

Biological Interpretation Boundary

Enzymatic activity observed in vitro or biological effects observed in experimental models do not establish human pharmacology, clinical safety, clinical efficacy or therapeutic benefit.

The biological rationale for D016 therefore remains a development hypothesis that must be tested progressively through appropriate preclinical and, where justified, potential future clinical development.

D016 connects a defined recombinant enzyme design with a broader scientific question: whether its designed PH20-like and HYAL4-like biochemical profile can generate biologically relevant effects that justify further pharmaceutical development.

Evidence – not the biochemical rationale alone – determines whether that development hypothesis should progress.

Pharmaceutical Development

From D016 Technology to BELACT® Development

D016 provides the recombinant hyaluronidase technology underlying BELACT®, Pantarhin’s lead investigational pharmaceutical development program.

BELACT® translates selected scientific and biological questions associated with D016 into a structured pharmaceutical-development framework in which indication strategy, preclinical evidence, pharmaceutical readiness, regulatory requirements and potential clinical development are evaluated separately.

D016 Technology

D016 defines the underlying recombinant enzyme technology, including its biochemical characteristics, designed PH20-like and HYAL4-like activity profile and associated analytical characterization.

Technology characteristics alone do not define an indication, dose, clinical regimen or therapeutic claim.

BELACT® Development Program

BELACT® is the lead investigational pharmaceutical development program based on D016 and provides the framework through which potential therapeutic applications of the technology are evaluated.

BELACT® is therefore a development program designation and should not be interpreted as an approved medicinal product or as synonymous with D016 itself.

Focused HoFH Development Path

Within BELACT®, homozygous familial hypercholesterolemia (HoFH) is being evaluated as the planned lead indication for a focused orphan-oriented development path.

HoFH has not been approved as an indication for BELACT®, and no orphan-drug designation or other expedited U.S. regulatory designation has been granted.

Broader Cardiovascular Development

Pantarhin also evaluates whether the D016-related scientific rationale and its U.S. therapeutic-use intellectual-property position may support broader cardiovascular development opportunities beyond the planned HoFH entry path.

Any broader cardiovascular indication would require its own scientific rationale, evidence package, development strategy, regulatory assessment and resource commitment.

Evidence-Gated Progression

Progression from technology characterization toward pharmaceutical and potential clinical development is not automatic. Each stage requires evidence and readiness appropriate to the next development decision.

Scientific findings, preclinical results, pharmaceutical-development status, manufacturing readiness, regulatory input, ethical requirements and available resources may support progression, modification, additional investigation, hold or discontinuation.

Regulatory Strategy

Regulatory interactions are used to inform the development requirements applicable to BELACT® and any future pharmaceutical development based on D016.

Such interactions may clarify development expectations but do not constitute approval, endorsement, authorization, designation or confirmation of clinical safety or efficacy.

Distinct Development Layers

D016 technology, the BELACT® development program, individual potential indications and Pantarhin’s U.S. therapeutic-use patents represent related but legally, scientifically and developmentally distinct layers.

Reference to one layer should not be interpreted as establishing rights, evidence, regulatory status or development conclusions for another.

Rights Context

Participation in BELACT® development or access to D016-related scientific information does not by itself establish ownership of D016, unrestricted development rights, licensing authority or commercialization rights.

Applicable rights and responsibilities are determined by the relevant corporate, intellectual-property and contractual documentation.

D016 provides the technological starting point; BELACT® provides the pharmaceutical-development framework through which selected therapeutic hypotheses are evaluated.

Whether any development path advances depends on the evidence and readiness generated within that framework – not on the existence of the technology, program designation or intellectual-property position alone.

Nothing on this page establishes an approved indication, clinical efficacy, therapeutic benefit, regulatory designation or authorization for BELACT®, D016 or any future pharmaceutical product based on D016.

Such access does not by itself transfer manufacturing technology, intellectual-property ownership, unrestricted development rights, licensing authority, exclusivity or commercialization rights.

Separate Intellectual-Property Layers

D016-related technology rights and Pantarhin’s issued U.S. therapeutic-use patents represent distinct intellectual-property layers.

Pantarhin’s therapeutic-use patents may be relevant to defined cardiovascular development applications involving hyaluronidase, but they should not be interpreted as ownership of the underlying D016 technology or as unrestricted rights across all potential uses of D016.

Licensing and Commercial Rights

Any licensing, co-development, supply, territorial, field-of-use or commercialization rights must be defined specifically within the applicable rights and contractual framework.

The ability to participate in scientific or pharmaceutical development does not automatically establish authority to grant rights in D016 or in intellectual property owned or controlled by another entity.

Rights and disclosures are defined by agreement – not inferred from access to material or information.

Development Discussions

Pantarhin welcomes relevant scientific, pharmaceutical-development and strategic inquiries concerning D016-related development activities within the applicable rights framework.

Additional technical or non-public information may be considered separately where the purpose of the inquiry, confidentiality requirements and applicable rights support further discussion.

D016 remains an investigational technology. Nothing on this website establishes ownership, unrestricted development or commercialization rights, an approved pharmaceutical product, an approved indication, clinical safety, clinical efficacy or therapeutic benefit.

Any future pharmaceutical development remains subject to appropriate scientific evidence, pharmaceutical readiness, regulatory and ethical requirements, applicable rights and available resources.

D016 is therefore presented by Pantarhin as a defined recombinant hyaluronidase technology within a controlled pharmaceutical-development framework – with scientific evidence, development responsibilities and legal rights evaluated as separate but interdependent elements.

Development proceeds where evidence and defined rights support it.
